The test medicine will be 'radiolabelled' - it will contain a small amount of radioactivity (Carbon-14) - so that it can be traced in the body.\n\nParticipants will take part in both study parts and will remain resident in the clinic for up to 10 days after their final dose of Investigation Medicinal Product (IMP). After the participants have left the clinic, they may have to collect their urine and faeces at home until mass balance criteria has been met.\n\nOver a period of at least 8 days in Part 1 and 10 days in Part 2, many blood samples will be taken, and volunteers will collect all their urine and faeces so that the amount of test medicine and its breakdown products can be measured\u002Fanalysed.\n\nA blood sample will be taken from volunteers between admission and first dose for pharmacogenetic testing.",[57,63,68],{"label":58,"type":59,"description":60,"interventionNames":61},"Ontunisertib","EXPERIMENTAL","Participants will receive single oral dose of ontunisertib",[62],"Drug: Ontunisertib",{"label":64,"type":59,"description":65,"interventionNames":66},"[14C]ontunisertib solution for infusion","Participants will receive single IV dose of \\[14C\\]ontunisertib",[67],"Drug: [14C]ontunisertib",{"label":69,"type":59,"description":70,"interventionNames":71},"[14C]ontunisertib","Participants will receive single oral dose of \\[14C\\]ontunisertib",[67],[73,79,82],{"type":74,"name":58,"description":75,"armGroupLabels":76,"otherNames":77},"DRUG","Oral capsule",[58],[78],"AGMB-129",{"type":74,"name":69,"description":80,"armGroupLabels":81,"otherNames":47},"IV infusion",[64],{"type":74,"name":69,"description":75,"armGroupLabels":83,"otherNames":47},[69],[85],{"name":86,"affiliation":87,"role":88},"Silke Hüttner, MD","Agomab Therapeutics","STUDY_DIRECTOR",[90],{"name":91,"role":92,"phone":93,"phoneExt":47,"email":94},"Agomab Clinical Operations","CONTACT","0032 3318 91 70","clinicalstudies@agomab.com",[96],{"facility":97,"status":98,"city":99,"state":47,"zip":100,"country":101,"countryCode":102,"cosmosGeoPoint":103,"geoPoint":108,"contacts":47},"Quotient Sciences Limited","RECRUITING","Nottingham","NG11 6JS","United Kingdom","UK",{"type":104,"coordinates":105},"Point",[106,107],-1.15047,52.9536,{"lat":107,"lon":106},{"type":110,"investigatorFullName":47,"investigatorTitle":47,"investigatorAffiliation":47,"oldNameTitle":47,"oldOrganization":47},"SPONSOR","100644825","phase-1-human-mass-balance-study-of-14c-ontunisertib-in-healthy-volunteers-100644825",false,"NCT07672574","Human Mass Balance Study of [14C] Ontunisertib in Healthy Volunteers","An Open-label, Two-part Trial to Assess the Mass-balance Recovery, Metabolite Profile, Metabolite Identification and Absolute Bioavailability of [14C] Ontunisertib in Healthy Male Participants","Inclusion Criteria:\n\n* Males aged 30 to 65 years inclusive at the time of signing informed consent\n* Must agree to the use of an adequate method of contraception for up to 93 days post-final dose\n* Healthy male participants according to the assessment of the investigator, as based on a complete medical history including a physical examination, vital signs, 12-lead ECG, and laboratory safety tests without any clinically significant abnormalities.\n* Body mass index of 18.0 to 29.9 kg\u002Fm2\n\nExclusion Criteria:\n\n* Serious adverse reaction or serious hypersensitivity to any drug or formulation excipients\n* History of clinically significant renal, hepatic, respiratory, particularly GI disease, especially peptic ulceration, clinically significant GI bleeding, ulcerative colitis, Crohn's Disease or Irritable Bowel Syndrome, and neurological or psychiatric disorder\n* History of GI surgery (with the exception of appendectomy or hernia repair unless it was performed within the previous 12 months)\n* Clinically significant abnormal clinical chemistry, hematology, coagulation or urinalysis as judged by the investigator at screening.\n* Radiation exposure, including that from the present trial, excluding background radiation but including diagnostic x-rays and other medical exposures, exceeding 5 mSv in the last 12 months or 10 mSv in the last 5 years.
