About this trial

The RESET-INSPIRE study is designed to investigate individual variability in glucose response and insulin sensitivity, a key mechanism underlying metabolic-associated steatotic liver disease (MASLD) and cardiometabolic disorders. Building on findings from Project RESET, this study aims to develop and validate a continuous glucose monitoring (CGM)-based approach for assessing insulin sensitivity.

Eligibility criteria

This trial accepts healthy volunteers

Qualifiers

Adults aged between 21 and 70 years (including 70 yrs old) at the time of screening

Able to speak and understand English

No past history of acute myocardial infarction (heart attack), stroke or heart failure

Able to understand and voluntarily provide written informed consent prior to any study specific procedures.

Disqualifiers

Known diagnosis of diabetes mellitus (type 1 or type 2), or use of antidiabetic medication.

Current or recent (within 3 months) use of medications known to affect glucose, including but not limited to corticosteroids.

Known history of liver disease or abnormal liver function deemed clinically significant by the investigator who is a medical doctor.

Renal impairment, defined as eGFR ≤ 60mL/min/1.73m²

Trial population

Recruitment is in Singapore as follows: 1. Flyer: The flyer, which contains a QR code to our landing page and the study contact number, will be distributed at various outreach events such as the Standard Chartered Marathon Race Pack Collection. Permissions will be obtained from the event organizers (if necessary, especially for non-NUS events), before distribution of flyers. 2. Word of Mouth: We are planning to inform participants of Project RESET about this study and encourage them to inform their family and friends to participate in it. 3. Project RESET Cohort: About 5000 people have signed up for Project RESET and we can look to recruit these people for this study if they have given their consent to be contacted for other studies. 4. Expression of interest online form: There was an online Town Hall conducted for all Project RESET participants on 11th and 12th of February 2026 and Project INSPIRE was introduced there with a link and QR code to Project INSPIRE.

15 days
2

Blood Glucose

Blood glucose is measured at all timepoints for the OGTT and is output in mmol/L

Time frame
2 hours
3

Insulin

Insulin in the blood is measured at all timepoints of the OGTT and output in mU/L

Time frame
2 hours
4

C-Peptide

C-Peptide levels in the blood is measured at all timepoints of the OGTT and output as ng/ml

Time frame
2 hours

Secondary outcomes

1

HBA1C

Blood HBA1C measured in the fasting blood sample during the OGTT and output in percentage (%)

Time frame
At the start of OGTT
2

hs-CRP

hs-CRP measured in the fasting blood sample and output as mg/L.

Time frame
At the start of the OGTT
3

ESR (Alifax)

Erythrocyte Sedimentation Rate (Blood) measures ESR (Alifax) and outputs in mm.hr

Time frame
At the start of OGTT
